CYCLOGESTON is a monthly intramuscular injectable contraceptive consists of a mixture of a long acting progestogen and a long acting estrogen. The depot effect results from the slow absorption from the injection site.
Parenterally, medroxyprogesterone acetate is a long acting progestogen and a long acting progestational steroid. Medroxyprogesterone acetate administered parenterally to women with adequate endogenous estrogen transforms proliferative endometrium into secretory endometrium.
Medroxyprogesterone inhibits the secretion of pituitary gonadotropin which, in turn, prevents the follicular maturation and ovulation. The effects on the endometrium, the fallopian tubes and progestational changes in the cervical mucus, inhibits sperm penetration into uterus, thus increasing the difficulty of nidation.
The progestin alone had been shown to interfere with fertility when administered intramuscularly. However, complete disruption of the menstrual cycle ensued. The long acting estradiol was added in the formula to induce monthly uterine bleeding mimicking the normal menstrual pattern as closely as possible.
